Pada penelitian ini digunakan jenis teh dari klon varietas Assamica, yakni teh Gambung di PT. Perkebunan Nusantara VIII Unit Sinumbra di Rancabali, Jawabarat. Teh Gambung diambil sampelnya pada musim kemarau menjelang musim hujan, dengan intensitas curah hujan yang rendah. Teh gambung yang ada pada PT. Perkebunan Nusantara VIII Unit Sinumbra ini termasuk dalam dataran tinggi. Citra daun teh diambil pada pencahayaan Lux 11.000 – 100.000 dari setiap blok.
